Precise mathematical descriptions of ligand–protein interactions are hindered by the inability to experimentally measure affinity and cooperativity. These parameters can be estimated from binding curves and agonist binding models. However the ability to confidently parameterize appropriate biophysical models is limited. We have analyzed the mathematical basis of the general problem of ligand binding and have developed general methods to determine whether the parameter values are well constrained.
